چکیده

Purpose: To investigate the neuroprotective effect of erythropoietin (EPO) against hypoxic-ischemic brain injury in neonatal mice, and underlying mechanism(s) action.
Methods: Sixty mice were assigned to 3 groups: sham, model EPO intervention groups, each with 20 mice. Western blot assay was used determine changes protein expressions AMPK, UCP2 KLF2. Fluorescence intensity reactive oxygen species (ROS) evaluated flow cytometry.
Results: The AMPK KLF2 cerebral tissues significantly up-regulated, relative (p < 0.05), while level also higher than 0.05). Mean fluorescence ROS cortex sham but it down-regulated group, group 0.05).
Conclusion: exerts by activating pathway, up-regulating expression UCP2, inhibiting production mitochondrial ROS, reducing oxidative stress tissue. Thus, has potentials for use clinical practice as a agent.

Neuroprotective effect of erythropoietin on hypoxic-ischemic brain injury in neonatal rats.

Erythropoietin (Epo) prevents ischemia and hypoxia-induced neuronal death in vitro. Recent studies have shown that this cytokine also has in vivo neuroprotective effects in cerebral and spinal ischemia in adult rodents. In this study, we aimed to investigate the effect of systemically administered recombinant human Epo on infarct volume and apoptotic neuronal death in a newborn rat hypoxic-isch...

C1q-deficiency is neuroprotective against hypoxic-ischemic brain injury in neonatal mice.

BACKGROUND AND PURPOSE This study was undertaken to determine whether the initial component of the classical complement (C) activation pathway contributes to hypoxic-ischemic brain injury in neonatal mice. METHODS Hypoxia-ischemia (HI) was produced in C1q(-/-) and wild-type (WT) neonatal mice. At 24 hours after HI, neonatal mouse reflex performance and cerebral infarct volume were assessed. L...

Neuroprotective agents for neonatal hypoxic-ischemic brain injury.

Hypoxic-ischemic (H-I) brain injury in newborns is a major cause of morbidity and mortality that claims thousands of lives each year. In this review, we summarize the promising neuroprotective agents tested on animal models and pilot clinical studies of neonatal H-I brain injury according to the different phases of the disease. These agents target various phases of injury including the early ph...

The neuroprotective effect of recombinant human erythropoietin via an antiapoptotic mechanism on hypoxic-ischemic brain injury in neonatal rats

PURPOSE The neuroprotective effects of erythropoietin (EPO) have been recently shown in many animal models of brain injury, including hypoxic-ischemic (HI) encephalopathy, trauma, and excitotoxicity; however, limited data are available for such effects during the neonatal periods. Therefore, we investigated whether recombinant human EPO (rHuEPO) can protect against perinatal HI brain injury via...

Neuroprotection against hypoxic-ischemic brain injury by inhibiting the apoptotic protease activating factor-1 pathway.

BACKGROUND AND PURPOSE Emerging evidence suggests that mitochondrial damage-mediated neuronal apoptosis is a major contributor to neonatal hypoxic-ischemic (H-I) brain injury. This study was performed to determine whether targeted inhibition of the apoptotic protease activating factor-1 (Apaf-1) signaling pathway downstream of mitochondrial damage confers neuroprotection in rodent models of neo...
